Abstract
An High-performance liquid chromatographic method with tandem mass spectrometry (HPLC-MS/MS) was developed for the determination of Chloramphenicol (CAP) residue in fish.The CAP residue was extracted by ethyl acetate.Concentrated extracts were redissolved in 1 mL dissolving solution and defatted by hexane.The solution was separated on a RP -HPLC column and detected by HPLC-MS/MS with positive electrospray ionization(ESI-) in multiple reaction monitoring(MRM) mode.Quantitation was carried out using an external standard method.The standard curve for CAP was linear over the concentration range of 0.04 ~1.0 μg/kg with correlation coefficients (R2) of 0.9999. The recoveries of CAP from fish samples spiked with the concentration levels at 0.10, 0.20, 0.40 μg/kg were range of 91.0%~100.1%. The relative standard deviations were 2.6%~4.1%. The limit of detection for the method was 0.01 μg/kg. The limit of quantification for the method was 0.02 μg/kg.
