Abstract

Objective To investigate the relationship between GABAA receptor and the amnesic effect of propofol. MethodsThe mice were randomly divided into 5 groups (n=10 each): I group (intralipid 10 ml·kg-1 group), P25+NS group (propofol 25 mg·kg-1 +normal saline 10 ml·kg-1 group), P25+Se1 group (propofol 25 mg·kg-1 +securinine 1 mg·kg-1 group), P25+Se2 group (propofol 25 mg·kg-1+securinine 2 mg·kg-1 group), and P25+Se4 group (propofol 25 mg·kg-1 +securinine 4 mg·kg-1 group). The step-through/step-down latency and the number of errors were recorded 24 h after the drugs were given in the tests. ResultsAs the P25+Se2 group and the P25+Se4 group were compared with the P25+NS group,the step-through latency and the step-through latency prolonged and the numbers of errors decreased(P0.01),seemingly in a dose-dependent manner; but none of them attained the levels of I group. ConclusionGABAA receptor partially mediates the propofol-induced amnesia in mice.
