Abstract
Objective To observe the effects of TNF-α on the TF and TFPI antigen expression in HUVECs. Methods HUVECs were cultured to TNF-α (10 ng / mL) for the six various lenghts of time(0 h,1 h,3 h,6 h,12 h,24 h),and the various concentration of TNF-α (0,1,10,20,50,100 ng / mL) for 6 h. TF and TFPI antigen were determined by ELISA; TF mRNA and TFPI mRNA were examined by RT-PCR. Results There were no significant difference in cell viability between treated groups and untreated groups; TNF-α increased TF antigen expression in HUVECs in a dose or time-dependent manner,reaching a maximum level with TNF-α 10 ng / ml after 6 hours (P 0.01); TNF-α increased TF mRNA expression in HUVECs in a dose or time-dependent manner,reaching a maximum level with TNF-α 10 ng / mL after 3 hours (P 0.01); There was no effect on the TFPI antigen and mRNA exprssion induced by TNF-α in HUVECs. Conclusions TNF-α can induce the TF expression in HUVECs; There has no effect on TFPI expression induced by TNF-α in HUVECs.
