【Objective】 The objective of the experiment is to understand the diversity and genetic structure of oriental persimmon(Diospyros kaki) populations in Shandong.【Method】 The genetic diversity of 207 individuals from 8 persimmon(Diospyros kaki) populations in Shandong were investigated using SSR markers.The genetic parameters were analyzed using POPGENE 1.31 software,genetic variation of interpopulation and inner-population were analyzed by AREQUIN 2.0 version,and then the mean expected heterozygosity(He) and genetic differentiation coefficient were counted by TFPGA software.【Result】 As a result,128 bands,of which 105 were polymorphic loci,were amplified by 10 SSR primers selected from 51 markers.The relatively high levels of PPL(82%),Nei's gene diversity(Heranged from 0.153 8 to 0.247 6) and Shannon’s information diversity(H0ranged from 0.221 7 to 0.372 1) revealed relatively rich genetic diversity in the 8 persimmon populations.Analysis of molecular variance(AMOVA) showed that the variance component among populations contributes 24.17% at a significantly high level(P0.001).Furthermore,a low level of genetic variation among populations was detected based on the genetic distance(D 0.231 7),identity(I 0.783 2).Nei's genetic diversity analysis and AMOVA revealed that there was a higher level of genetic differentiation among populations(φST= 0.222 6,FST= 0.175 9).【Conclusion】The oriental persimmon populations in Shandong possess abundant genetic diversity,the genetic differentiation between populations may result from human selective pressures.
