Abstract
Objective To investigate the anti-proliferation effect of curcumin and curcumin combined with cisplatin on human ovarian cancer cell line SKOV-3 in vitro.Methods Human ovarian cancer SKOV-3 cells were cultured and treated with different concentrations of curcumin(5,10,20,40 and 80 μmol/L),cisplatin(10 μmol/L),or curcumin combined with cisplatin in different groups.The proliferation of SKOV-3 cells was detected by MTT assay.Flow cytometry was used to analyze the apoptosis and cell cycle distribution.Results MTT assay showed that curcumin of different concentrations could significantly inhibit the proliferation of SKOV-3 cells in time-dependent and dose-dependent manners(P0.05).Compared with that in single drug groups,the suppression in combination treatment group was significantly higher(P0.05).Flow cytometry showed that curcumin could arrest SKOV-3 cells in G2/M phase.Furthermore,curcumin could induce apoptosis.With the increased concentration of curcumin treatment,the apoptosis rate increased.Conclusion Curcumin can inhibit the growth of SKOV-3 cells.In a certain range,the effect is in dose-and time-dependent manners.With curcumin combined with cisplatin,the effect could be enhanced in a synergistic manner.
