Effects of Methyl Jasmonate on the Accumulation of Phenolic Acids in Salvia miltiorrhiza Hairy Root

Abstract

OBJECTIVE To study the effects of methyl jasmonate (MJ) on the accumulation of the main water-soluble phenolic acids in Salvia miltiorrhiza hairy roots. METHODS The different dosages of MJ were added into the suspension cultures at different growth stages of S. miltiorrhiza hairy roots. The contents of Danshensu,rosmarinic acid,and salvianolic acid B were determined by HPLC. RESULTS MJ affected the accumulation of these compounds in a dose-dependent manner. The MJ adding time also showed significant influence on the accumulation. The peak content of Danshensu was (0.510±0.054)% by 300 μmol·L-1 MJ cultured with the hariy root for 15 d. The peak contents of rosmarinic acid was (14.358±0.710)% by adding 200 μmol·L-1 MJ culture with the hariy root for 10 d. The peak content of salvianolic acid B was (1.592±0.120)% by 200 μmol·L-1 MJ cultured with the hariy root at the beginning. CONCLUSION MJ could obviously affect the accumulation of Danshensu,rosmarinic acid,salvianolic acid B,as well as the growth of the hairy roots. It is helpful for the further research of the biosynthesis of phenolic acids in Salvia miltiorrhiza Bunge.
