Protective effect of IL-10 on rats with Alzheimer′s disease induced by Aβ_(1-42)

Abstract

Objective To investigate the effect of IL-10 on expression of amyloid precursor protein(APP) in the hippocampus of rats with Alzheimer′s disease(AD) induced by Aβ1-42.MehtodsForty-eight rats were equally randomized into six groups of A(normal control),B(PBS control),C1(Aβ1-42 0.1 mM),C2(Aβ1-42 0.2 mM),C3(Aβ1-42 0.4 mM) and D(IL-10 80 μg/ml plus Aβ1-42 0.4 mM).Two days after injection,the latent period of rats was measured by Morris water maze,and the protein expression of APP in the hippocampus was detected by Western blot.Results Compared with groups of A and B,the latent period of rats in Morris water maze test and the protein expression of APP were increased in a dose-dependent manner in groups of C1,C2 and C3(P0.01).While the latent period and APP expression in group D were lower than those in group C3(P0.05).ConclusionIL-10 injected into the hippocampus can inhibit the increase of latent period and APP expression in the rats with AD,which indicates that IL-10 may depress the nerve inflammation of AD induced by Aβ1-42.
