EFFECT OF SEAWEED PIGMENT GLYCOPROTEIN ON EXPRESSIONS OF Caspase-3 AND Bax PROTEIN IN HEPATOMA CARCINOMA CELLS

Abstract

Objective To investigate the effect of seaweed pigment glycoprotein(SPG) on the expressions of Caspase-3 and Bax protein in liver cancer cells. Methods Fifty mice subcutaneously inoculated with liver cancer H22 cells were evenly randomized to five groups.The mice in high-,medium-and low-dosage SPG group were intragastrically administered with 100,50 and 10 mg/kg of SPG 0.5 mL once a day,respectively,for 10 days;those in the control group were given equal-volume of normal saline,once daily,for 10 days;those in cylophosphamide group were intraperitoneally injected with 20 mg/kg cyclophosphamide,every other day,for 10 days.All the mice were sacrificed the second day of the last medication,the cancer tissue of liver was taken.The proliferation activity of cancer cells was determined by MTT assay,and the expressions of Caspase-3 and Bax were measured by immunohistochemical method. Results The expression levels of Caspase-3 and Bax in high-dosage group were higher than that in the control group,and the proliferation activity was lower,the difference being significant(F=344.264-19 266.181,q=14.88-185.56,P0.05). Conclusion SPG can motivate the expressions of Caspase-3 and Bax protein in the cells of liver cancer,inducing the apoptosis of the cells.
