Abstract
Objective: To observe the high density lipoprotein(HDL) sub-fractions and anti-oxidation function of Rosiglitazone,and to further explore the potential anti-atherosclerosis mechanism of Rosiglitazone in rabbits.Methods: A total of 18 rabbits were randomly divided into three groups: Control group,the animals were fed with regular diet;Atherosclerosis group,the animals were fed with high cholesterol diet;and Rosiglitazone group,the animals were fed with high cholesterol diet plus Rosiglitazone 0.5mg/kg/day.n=6 in each group and all rabbits were treated for 12 weeks.Serum lipids were checked by enzymatic method and paraoxonase1(PON1) activity was assessed by spectrophotometry.HDL distribution was determined with chemical precipitation and the aorta tissue was observed by histological examination.Results: Compared with Atherosclerosis group,the serum level of HDL cholesterol(HDL-C),PON1 activity and the ratio of HDL2/HDL were significantly increased in Rosiglitazone group;the aortic intima-media thickness and the percentage of plaque area(surface area of plaque/surface area of whole intima) were significantly decreased in Rosiglitazone group.All the differences had the statistic meaning.Conclusion: Rosiglitazone could increase the serum level of HDL,PON1 activity,and the ratio of HDL2/HDL;decrease the aortic intima-media thickness and the percentage of plaque area,those activities might play the role of anti-atherosclerosis.
