Abstract
Objective To study the effects of ischemic postconditioning and adenosine postconditioning on lung ischemia-reperfusion injury in rats.Methods Rat models of right lung reperfusion iniury were induced by occluding hilum of right lung.Forty SD rates were randomly divided into four groups:sham operated group(n=10):continuous perfusion for 165 minutes;ischemic-reperfusion group(n=10):ischemia for 45 minutes and reperfusion for 120 minutes;adenosine postconditioning group(n=10):ischemia for 45 minutes,adenosine(1.5 mg·kg-1) bolus injection through right ventricle before reperfusion,then reperfusion for 120 minutes;ischemic postconditioning group(n=10):ischemia for 45 minutes,transient reperfusion for 10 seconds,ischemia for 10 seconds,which were repeated for 5 times,and then recovered to comprehensive reperfusion for 120 minutes.In the end of experiment,the lung tissue was taken to determine the wet/dry (W/D);the lung tissue structure was observed by using optics-scope.ELASA was used to determine the contents of IL-6 and IL-10.UV spectrophotometer was used to determine the content of MDA and SOD.Results Compared with ischemia-reperfusion group,ischemic postconditioning group and adenosine postconditioning group had similar effects:the wet-to-dry ratio decreased significantly(P0.05);content of SOD increased significantly(P0.01) and MDA decreased significantly(P0.05);content of IL-6 decreased and IL-10 increased(all P0.01).Conclusions Ischemic postconditioning and adenosine postconditioning can provide protective effects on lung ischemia-reperfusion injury by preventing lipid peroxidation and free radical formation,inhibiting proinflammatory cytokines IL-6 and increasing protective cytokine IL-10.
