Effect of Varicocele on Ultrastructure and Apoptosis in Rat Leydig Cells

Abstract

Objective:To study the effect of varicocele on ultrastructure and apoptosis in rat Leydig cells and to find mechanism of male infertility.Methods:The varicocele(VC)model was established by partial ligation of the left renal vein in male SD rats,and 20 rats were randomly divided into the VC group(n=10)and control group(n=10).All the animals were killed 90 dafter operation.The FSH,LH,and testosterone levels in blood of rats were detected by radioimmunoassays.Leydig cell ultrastructure of rats were observed with an electron microscope.Leydig cell apoptosis of rats was detected by flow cytometry.Results:Compared with the control group,FSH and LH levels of VC group were significantly higher(P0.05)while testosterone levels were significantly decreased(P0.05);Atrophy nucleichromatin condensation,perinuclear accumulation of chromatin,unclear nucleolus,and vacuolar dissolved cytoplasm were observed by electron microscopy in VC group.Leydig cell apoptosis rate of VC group was significantly higher than that of control group(P0.01).Conclusion:Varicocele could severely damage Leydig cells of rats,then induce disorders of sex hormone secretion in rats.
