Abstract
Objective To study the effect of arsenic trioxide on apoptosis of scar-derived fibroblast. Methods The scarderived fibroblasts were treated with arsenic trioxide in the doses of 0, 2, 4, 8 μmol/L for 0, 24, 48,72h. Their morphology and Bcl-2/Bax and caspase-3 expression were determined by Hoechst 33258/PI fluorescence staining and immunohistochemical method, respectively. Results The apoptotic bodies of fibroblasts and caspase-3 expression increased, but Bcl-2/Bax ratio decreased in dose-and time-depen-dent manner. Conclusion Arsenic trioxide may induce apoptosis of scar-derived fibroblasts by downregulation of Bcl-2/Bax ratio and overexpression of caspase-3.
