Association of TNF-α-850 SNP with Ankylosing Spondylitis

Abstract

Objective:To find the susceptible genes locus of ankylosing spondylitis in HLA region in Jilin Chinese population.Methods:Peripheral blood of 79 AS patients and 132 healthy subjects was collected.11 SNP loci in HLA region which were reported with strong linkage to other autoimmune diseases were selected and the frequencies of genotypes and allele distribution of single SNP were analyzed by case-control study.Results:The distribution of TNF-α-850 TT genotype was higher in AS group than in normal control group (P=0.027);Allele T had a significant statistical difference between AS group and normal control group(P=0.002).There was a very large difference of the positive ratio of HLA-B27 between two groups(χ2=155.264,P=0.000).After gene interaction analysis of HLA-B27and TNF-α-850,the RR was significantly higher in both TNF-α-850 T allele and HLA-B27 positive than HLA-B27 lonely.Conclusion:TNF-α-850 C→T mutation may increase the sicken risk of AS in jilin Chinese population.
