Age-related increase of β_3-adrenergic receptor in rats with experimental heart failure

Abstract

Objective To investigate the age-related expression of β3-adrenergic receptor (β3-AR) in rats with experimental heart failure.Methods The model of heart failure was established by banding the aortic diameter.Male Wistar rats were divided into young and aging groups according to age,and each group included sham-operation and heart-failure subgroups.At 0 and 9 weeks post-operation,all below were measured:left ventricular end-diastolic pressure (LVEDP),left ventricular end systolic pressure (LVESP),maximal rate of left ventricular pressure rise (dp/dtmax),and fall (dp/dtmin);myocardial β3-AR mRNA by RT-PCR.Results Left ventricular end-diastolic pressure (LVEDP) were significantly higher in the aged heart-failure group than in young heart-failure group (15.84±4.03 vs. 10.92±1.68,P0.01) and than the sham-operation groups.While the left ventricular end systolic pressure (LVESP) and the maximal rate of rise or fall of left ventricular pressure were all significantly lower in the aged heart-failure group than in the young heart-failure group and in the sham-operation groups (all P0.01).Myocardial β3-AR mRNA were significantly increased in the heart failure group compared to sham-operation group and was also significantly higher in the aged heart-failure group than in the young heart-failure group (0.78±0.05 vs. 0.65±0.09,P0.05). Conclusions These results suggest that there is an age-dependent upregulation of myocardial β3-AR mRNA in both sham-operation rats and in rats with experimental heart failure.β3-AR expression increased significantly when heart failure worsened in aging rats.
