The Effect of Tumor Necrosis Factor and Endotoxin in Severe Acute Pancreatitis

Abstract

Objective:To investigate the effect of tumor necrosis factor-α(TNF α) and endotoxin in the acute severe pancreatitis.Method:After SAP model induced in dogs,plasma levels of TNF α?endotoxin and amylase were measured at 6h?12h?24h?48h?after operation.Pathological changes and ultrastructure of pacreatitis were observed.Results:The plasm levels of TNF α?endotoxin and amylase were significantly higher in SAP group than in the control group,especially on 12h?24h?(P0.01~P0.05).The tissue demages of pancreas were observed.The pancreatic cells observed by microscope were demaged.There were hemorrhagic necrosis.Part of nucleus were karyolysis.There were much leukocyts around the necrosis tissue of pancreas.Conclusion:The TNF α and endotoxin were important factors in the development of severe acute pancreaetitis.The development of severe acute pancreaetitis may be alleviated by inhibitng the production of TNF α and endotoxin.
