Abstract

Objective To investigate the effect of NCS-382 on the hypnotic and analgesic effects of isoflurane.Methods The mouse model of hypnosis was established and the model mice were divided into the control group and groups injected with different doses of NCS-382 (blocker of gamma hydroxybutyrate receptor).The effect of intrathecal (i.t.) injection of NCS-382 on the sleeping time (ST) was evaluated.In hot-plate test and acetic acid-induced writhing test,the mice were divided into a conscious group and an analgesic group;the two groups were further divided into control group and groups injected with different doses of NCS-382.The effects of intrathecal (i.t.) injection of NCS-382 on the pain threshold in hot-plate test (HPPT) and writhing times were assessed.Results Injection of 115,23,4.6,2.3,and 1.15 μg NCS-382 (i.c.v.) decreased ST of the mice treated with isoflurane (P0.01),while 0.92,0.184,and 0.036 8 μg NCS-382 had no significant effect on ST of mice (P0.05).Injection of 115,4.6,and 0.184 μg NCS-382 (i.t.) did not affect the HPPT and writhing times of conscious mice and the mice treated with isoflurane (P0.05).Conclusion GHB receptor may be one of the important targets for the hypnotic effects of isoflurane,and it may not be the main target for the analgesic effects.(Shanghai Med J,2010,33:319-321)
