Detection of the Cytotoxicity and Genotoxicity of Kuaishaling on Mouse Bone Marrow

Abstract

Objective To detect whether Kuaishaling has cytotoxicity and mutagenicity.Methods The agent was administrated to mice at some different doses (1/4×,1/2×,1×and 2×LD 50 )by gavage once a day for five days,then the mitotic index,the rate of chromosome structural aberrations and the rate of the cells with chromosome aberrations were detected by the methods of cytogenetics.Results Compared with the negative control group,Kuaishaling at the dose of 18.2 mg/kg could dramatically decrease the mitotic index of mouse marrow cells (P0.001);the rates of chromosomal structural aberrations and of the cells with aberrations were increased slightly at the dose of 18.2 mg/kg and 36.4 mg/kg and there was no significant differences between these two groups (P0.05),but these rates were increased significantly at the dose of 72.8 mg/kg and 145.6 mg/kg (P0.01).Conclusions kuaishaling can not only inhibit proliferation of bone marrow cells, but also cause chromosome damage, and the effection of the former is stronger than that of the later.
