Comparison of toxicities of breviscapine in Beagle dogs and monkeys

Abstract

Objective:The toxicities of intravenous breviscapine were compared in Beagle dogs and monkeys.Methods:In 32 Beagle dogs and 4 monkeys of either sex,breviscapine (at 40,80 and 120 mg·kg~(-1) in dogs;200 mg·kg~(-1) in monkeys) was intravenously dripped once daily for 13 weeks,and then recovered for 4 weeks.The toxic responses were observed.Results:In some Beagle dogs treated with 80 and 120 mg·kg~(-1)·d~(-1),vomiting,diarrhea and salivation were observed.At 11th day,one dog with 120 mg·kg~(-1)·d~(-1) showed macular rash and urticaria;one dog with 120 mg·kg~(-1)·d~(-1) died of he- matemesis and hematochezia.Only one monkey with 200 mg·kg~(-1)·d~(-1) showed an intermittent sialorrhea. The body weight,food consumption,hematological and biochemical variables in breviscapine groups were not different from those in control dogs and monkeys.Conclusion:The tolerance,sensitivity and toxicity of breviscapine were obviously different between Beagle dogs and monkeys.
