Effects of Lipopolysaccharide on Cytokines and Function of HPA-axis of Streptozocin-induced Diabetic Rats

Abstract

Objective To investigate the effects of lipopolysaccharide(LPS)on cytokines and functions of HPA axis of streptozocin-induced diabetic rats.Methods The experimental diabetic animal model was induced by streptozocin(50mg/kg),after that,the endotoxemia condition was induced by LPS [0.5mg/(kg bw)].The serum levels of cytokines such as TNF-α、IL-6 and plasma CRH、ACTH、corticosterone were evaluated at the spots of before LPS ip and after LPS ip 6、12、24、72h,respectively.Results The levels of TNF-α、IL-6 were significantly increased,and the peaks of TNF-α、IL-6 arrived at the spot of 12h after injection LPS.The plasma levels of CRH、ACTH and corticosterone were increased but there were no statistically significant to be observed between DM control and DM+LPS subgroups.There were no significant influences on glycemia and insulin levels to be observed in diabetic rats after LPS ip.Conclusion LPS-induced endotoxemia will increase the serum levels of TNF-α、IL-6 and activated the function of HPA axis.
