Abstract

Objective: To observe the effects of cigarette smoke extract(CSE) on the activation of cells and levels of tumor necrosis factor-α(TNF-α) in pulmonaryⅡepithelial cells(A549).Methods: A549 were cultured in vitro and exposed to different concentrations of CSE.Then the morphology and activity of the cells and levels of tumor necrosis factor-α(TNF-α)were observed by converted microscope,MTT and ELISA,respectively.Result:(1)The morphology and structure of A549 were changed obviously with different concentrations of CSE by different time.(2)The MTT in CSE groups were significantly decreased than that of the control one(P0.01).(3) The levels of TNF-α were in a time-dependent manner.The groups of 5% and 10% CSE were increased,while the 20% CSE group were decreased than that of the control group(P0.01).Conclusions: CSE could damage the morphology and activity of A549.The levels of TNF-α were changed by the injury.
