Fluconazole Inducing Penicillium Marneffei Mycelial Form to Produce 67.5-Kilodalton Protein

Abstract

Objective To investigate the change of exocrine protein during penicillium marneffei mycelial form cultured with fluconazole in vitro.Methods Eleven strains of penicillium marneffei were respectively cultured in 30 mL Sabouraud dextrose broth(SDB) with fluconazole concentration of 8 μg/mL and without fluconazole at 25℃ for one week.The MIC of fluconazole against mycelial fungi in different liquid culture were tested by E-test method.The proteins in liquid culture were extracted and secreted products were analyzed by polyacrylamide gel electrophoresis.Results The suspension of mycelial penicillium marneffei cultured in SDB with fluconazole presented rose color,but the suspension of mycelia cultured in SDB without fluconazole not presented rose color.The MIC of fluconazole against mycelia cultured with fluconazole increased significantly(P0.01) and furthermore they secreted 67.5-kilodalton protein.Conclusion Fluconazole concentration of 8 μg/mL can induce penicillium marneffei to secret red pigment and 67.5-kilodalton protein in SDB at 25℃.
