Abstract
Objective To find out an effective method for amplification and purification of dendritic cells(DCs) from peripheral blood of patients with chronic hepatitis B.Methods Peripheral blood monocytes were purified from peripheral blood of health volunteers(control group,10 cases) and patients with chronic hepatitis B(experimental group,10 cases) with incubation of granulocyte/macrophage colony stimulating factor(GM-CSF) and interleukin-4(IL-4).DC's quantity was counted under the inverted microscope and the expression levels of CD40,CD80,CD86 and MHC-DR on DCs were analyzed by flow cytometer after and before DCs incubation with GM-CSF and IL-4.Results The expression levels of CD40,CD80,CD86 and MHC-DR of DCs in the experimental group were significantly less than those in the control group(P0.01).DCs in the experimental group was significantly proliferated in the presence of GM-CSF and IL-4(P0.01).On the 5th day of the experiment,the expression levels of CD40,CD80,CD86 and MHC-DR of DCs in the experimental group were significantly increased(P0.01) and there was no difference versus the control group(P0.05).Conclusion The results suggest that combination of GM-CSF and IL-4 can effectively enhance proliferation and immune function of DCs from peripheral blood of patient with chronic hepatitis B.
