Abstract

The Mango leaf blight pathogen was identified as Pestalotiopsis mangiferae(P.Henn.) Steyaert.In vitro culture showed that Richard's medium supplemented with the boiled juice of mango leaves was the best for the mycelial growth of the pathogen.No conidium was produced when growing in Czapek's medium.The optimum temperature and pH value for the growth of hypha were 20~25 ℃ and pH 5.0~6.0.Sucrose and D-Glucose were the best carbon sources for mycelial growth while lactose and sucrose were the best for conidium germination.Sodium nitrate and Potassium nitrate are the best nitrogen sources for mycelial growth while ammonium nitrate was the best for conidium germination.Continuous light or 12 h light/ 12 h dark cycles were the best for Mycelial growth and conidium.Hypha growth was not good and spore formation was inhibited when cultured under complete dark.The lethal temperature for conidium was 52 ℃,10 min.
