In Vitro Stimulating Effect of Dendritic Cells Derived from Peripheral Blood Mononuclear Cells of Patients with Chronic Myelocytic Leukemia on Proliferation of T Cells

Abstract

Objective To induce dendritic cells (DCs) from peripheral blood mononuclear cells(PBMCs) of patients with chronic myelocytic leukemia(CML) ,observe the morphology and phenotype of the induced DCs,and study their stimulating effect on the proliferation of T cells.Methods Induce the DCs with GM CSF,IL 4 and TNF α in vitro,observe the morphology of induced DCs under inversed microscope and electromicroscope,and detect the phenotype by FACS.Detect the induced DCs for bcr/abl fusion gene by D FISH,and study their stimulating effect on the proliferation of T cells by MTT test.Results The bcr/abl fusion gene positive DCs (CML DCs) were induced from the PBMCs of patients with CML.The CML DCs showed significant stimulating effect on the proliferation of antogenic T cells,but CML showed no the effect.Compared with that of normal DCs,the stimulating effect of CML DCs on allogenic T cells was weak.However,after PBMCs were cultivated with 300 U/ml IFN α,the stimulating effect of induced DCs was similar to that of normal DCs.Conclusion CML DCs showed stimulating effect on the proliferation of autogenic and heterogenous T cells.However,the effect on allogenic T cells was weak compared with that of normal DCs.IFN α improved the stimulating effect of CML DCs.
