Abstract
The cells was cultured in 7?L bioreactor with the microcarrier of 3 g/L,dissolved oxygen of 50% and stirring speed of 40—50?r/min.The perfusion was performed after 48?h culture.The dynamic glucose concentration,fluctuations of osmotic pressure and pH value was monitored continuously.Meanwhile,the cell and proliferation were observed.Three consecutive batches in 7?L perfusion culture demonstrated high,quick cell proliferation speed,good cell adhesions and appearances,and cellular population doubling level reached 4.7.The perfusion culture approach in bioreactor is a quick way to prepare cells in large scale.
