Abstract
Objective To investigate the apoptosis and autophagy inducing effect of Cisplatin on HeLa cells.Methods MTT assay was employed to examine the growth suppression of the cells.Hoechst33342 staining was used to observe the apoptosis.Indirect immunofluorescence was used to detect the expressions and location of the LC3 and P62 proteins.Western Blotting was used to detect the expressions of Bax/Bcl-2,Caspase-3,LC3 and P62.Results The inhibitory effect of Cisplatin was in a time and dosage dependant manner.Cisplatin could increase the ratio of Bax/Bcl-2 and expression of caspase-3.Cispaltin treatment induced activation of LC3 and P62.Conclusion Cisplatin could induce apoptosis of HeLa cells paralleled autophagy.
