Abstract

OBJECTIVE To investigate the effects of tanshinone on rat vascular smooth muscle cell(VSMC) proliferation and ERK signaling METHODS Rat dorsal aortic VSMCs were cultured by tissue sticking method MTT and protein assay were used to estimate the proliferation activity of VSMC Western blot was used to assess the protein expression of pp ERK1/2 RESULTS Tanshinone inhibited VSMC proliferation response stimulated by 5% fetal bovine serum (FBS) and serum stimulated pp ERK1/2 expression in a dose dependent fashion Inhibition rates of 2,10,50 μg·mL -1 tanshinone were (17 7±3 6)%,(26 2±4 1)% and (72 7±3 8)%( P 0 05 vs control), respectively CONCLUSION The results suggested that tanshinone down regulated the expression of pp ERK1/2, which might be the mechanism in depressing VSMC proliferation
