Experimental Study on CD55,CD59 in Red Blood Cell and IL-8,IL-12 in Plasma of Liver Cancer Patient

Abstract
Purpose To study the relationship between CD55,CD59 in red blood cell and IL-8,IL-12 in patients with primry hepatocarcinoma (PHC) by the nature and isolation experimental method. Metheds Whole blood cells nature test tube (0.2ml whole blood cells suspension+0.3ml plasma+0.2ml normal saline) and white blood cells isolation test tube (0.2ml white blood cells suspension+0.3ml plasma+0.2ml normal saline) were incubated for 1h at 37℃. The content of IL-8 and IL-12 was detected by enzyme linked immunadsorbent assay (ELISA) method,and the expression of CD55 and CD59 in red blood cells was detected by flow cytometry. Results IL-8 in nature group and isolation group was (353.64±271.92)pg/ml and (424.97±278.93)pg/ml respectively in patient with PHC; (4.98±4.15)pg/ml and (20.72±14.20)pg/ml respectively in normal controls. IL-12 in nature group and islation group was (9.95±3.49)pg/ml and (13.02±3.74)pg/ml respectively in patient with PHC; (9.84±2.23)pg/ml and (13.59±3.69)pg/ml respectively in normal controls. There was positive correlation between CD59 in red blood cells and IL-8 in normal controls (r=0.669,P0.05). While in patient with PHC,CD55 in red blood cells were negative correlated with IL-8 (r=-0.727,P0.01). Conclusion In systemic hematogenic immunity,red blood cells regulate the level of IL-8 and IL-12 in plasma,but the capatity of red blood cells regulating IL-8 and IL-12 in patients with PHC was much lower than that in normal controls. In patients with PHC,content of CD55,CD59 in red blood cells and IL-8 in plasma are being unbalanced.
