The effect of high intensity ultrasound on the Th1/Th2 splenocyte of mouse bearing U14 cervical cancer cells

Abstract

Objective To investigate the effect of high intensity ultrasound on the T helper type 1 (Th1) and T helper type 2 (Th2) splenocyte subgroups of mouse bearing U14 cervical cancer cells. Methods Thirty-two mice were divided into 4 groups: a HIU group (A), a surgery resection group (B), a non-treatment group (C) and a normal group (D). Groups A, B and C were inoculated with the U14 cervical cancer cells, while the group D with normal saline. On the seventh day after tumor transplantation, the mice in the 4 groups received different therapy respectively; On the 17th day after tumor transplantation, the expression of cytokine was tested by enzyme-linked immunosorbent assay (ELISA) in splenocyte culture supernatants which was collected in every group. Results When splenocyte cells were stimulated by Con A, the IL-2 concentration in group A was significantly higher than that in group B (1110. 13±338. 10 pg/ml versus 641.50±93.29 pg/ml and 215.75±62.54 pg/ml, respectively) ( P0.05 ) ; the IFN-γconcentration in group A was sig- nificantly higher than that in group C (64984.86±9778.80 pg/ml versus 42613.75±4050.75 pg/ml) (P0.05 ) ; the IL- 4 concentration in group A was significantly lower than that in groups B and C (24.94±55.89 pg/ml versus 330.56±94.63 pg/ml and 418.13±104.78 pg/ml) (P0.05). When splenocyte cells were not stimulated by Con A, the IL-2 concentration in group A was higher than that in groups B, C and D ( 1099. 25±233.51 pg/ml versus 241.00±35.44 pg/ml, 136.75±37.52 pg/ml and 277.50±57.59 pg/ml) (P0.05 ) ; the IFN-γconcentration in group A (45390.00±9961.26 pg/ml) was significantly higher than that in group C (33662.50±14111.99 pg/ml ) ( PO. 05 ). Conclusion HIU-solidified cancer cells may activate the transformation of Th2 to Thl, regulate the balance of Th2 to Th1 type cytokine, play important role in improving immune function of tumor-bearing body.
