Abstract
Objective To explore a ability to inhibit apoptosis and low skeletal muscle atrophy.after denervation or not through the application of anti-oxidants grape seed.Methods Forty Vista rats randomly divided into five group: group A(control group),group B(14days denervated group),group C(28 days denervated group),group D(Intervention group 14 days denervation),group E(Intervention group 28 days denervation).Group A produced sham operation,Standard model of denervated gastrocnemius muscle was established by group B,C,D,E.After operation,group D,E were fed with grape seed 200mg·kg-1·d-1,group A,B,C were fed with normal saline.All rats were killed respectively in 14 days and 28 days.The content of reactive oxygen species(ROS) and the opening of the mitochondrial permeability transition pore(MPTP) in the gastrocnemius were detected respectively by flow cytometry(FCM) and fluorescence microscope;The apoptotic cells in atrophic muscle were detected by TUNEL.Results Compared with the group A after denervation,the expression of ROS,the opening of MPTP and the apoptosis of gastrocnemius were continued to increase(28 days) of group B,C,D,E.And the difference was significant(P0.05),Compared with the group B,C,the expression of ROS,the opening of MPTP and the apoptosis of gastrocnemius of group D,E were respectively decreased,the difference was significant(P0.05);The ratio of muscle wet weight of group B and C were significantly reduced than group A,The ratio of muscle wet weight of group D,E were respectively increased than group B and C.Conclusion ROS is a important element in regulating skeletal muscle atrophy after denervation,the grape seed can reduce the content of ROS,inhibit muscle cell apoptosis and ultimately delay skeletal muscle atrophy.
