A Study on Genotyping of Mycobacterium Tuberculosis and Its Drug Resistance

Abstract

Objective To study on genotyping of Mycobacterium tuberculosis(MTB) isolates and to analysis the relation between drug resistance and genotype of Variable number tandem repeat(VNTR)in Sichuan area.Methods Six tandem repeat loci of the 124 isolates of M.tuberculosis in Sichuan area were analyzed by PCR and agarose gel electrophoresis.The results of genotyping were analyzed with Gel-Pro analyzer 3.1 software and BioNumerics 3.0 software,and the relation between genetic type and drug resistance was explored.Results A total of 37 different allele profiles were identified by the VNTR for all the 124 isolates,74.19%(92/124)and 25.81%(32/124)belonged to cluster type and single type respectively.Drug resistant M.tuberculosis was significantly associated with cluster type.Conclusion There is obvious genetic polymorphism in the M.tuberculosis isolates in Sichuan area.And the clustering type was perhaps the main epidemic strains.It is suggested that surveillance of the epidemic genotype to be strengthened.
