Abstract
[Objective]To determine the contents of amylose and amylopectin in tobacco leaves.[Method]Dual wavelength spectrophotometry was used,and the measurement wavelengths of 462 and 610 nm for amylose,while 516 and 728 nm for amylopectin were selected according to the principle of dual wavelength colorimetric.The contents of amylose and amylopectin in tobacco leaves were calculated based on regression equation.[Result] The maximum absorption wavelength were 610 and 516 nm respectively for the two complexes,and the contents were 6.34% and 20.41%.[Conclusion] The contents of amylose and amylopectin in tobacco leaves determined with dual-wavelength spectrophotometry were accurate,and the method was suitable for the determination of amylose and amylopectin in tobacco leaves samples.
