Abstract

Objective To investigate the effect of Cinobufacini on on the proliferation inhibition and induction of apoptosis in glioblastoma cell line U251 and its molecular mechanisms. Methods Human glioblastoma(U251) cells were cultured in vitro.The growth inhibition of U251 cells was measured by clone formation assay.Western Blot analysis was performed to detect the expression of Bcl-2 and Bax. Results Clone formation test had shown that Cinobufacini inhibited significantly growth of U251 cells in a dose-dependent manner.Western Blot analysis showed that Bcl-2 expression were decreased,while Bax expression were increased after exposure to Cinobufacini.Conclusion Our results show that Cinobufacini strongly inhibited cell growth and proliferation human glioblastoma(U251) cells. Conclusions could significantly induce apoptosis of human glioblastoma cell U251;induction of apoptosis of Cinobufacini may be associated with down-regulation of the ratio of Bcl-2 / Bax.
