Abstract
Objective To evaluate the effects of edaravone postconditioning combined with remote isehemic postconditioning on myocardial ischemia-reperfusion(IR)injury in rats.Methods Sixty male Sprague-Dawley rats,aged 8weeks,weighing 250-300 g,were randomly divided into 4groups(n=15each):ischemia-reperfusion group(group C);edaravone postconditioning group(group E);remote ischemic pestconditioning group(group R);edaravone pestconditioning combined with remote ischemic pestconditioning group(group ER).Myocardial IR was induced by occlusion of left anterior descending artery for 30 min followed by 180 min reperfusion.Edaravone 5mg/kg was intravenously injected at 15 min before reperfusion in groups E and ER,and in groups C and R,0.5ml saline was intravenously injected.In groups R and ER,the animals underwent 10 min ischemia of bilateral hind limbs starting from 20 minutes after myocardial ischemia.At 180 min of reperfusion,the blood samples were taken from the jugular vein for determination of creatine kinase MB(CK-MB),malondialdehyde(MDA)andcardiac troponin I(cTnI)concentrations in serum.The animals were then sacrificed and myocardial infarct size was measured by Evans blue and TIC staining.Results Compared with those in group C,the myocardial infarct size(IS)and CK-MB,MDA and cTnI concentrations were significantly decreased in group E,group R and group ER(P0.05).Compared with those in group E and group R,the IS and the CK-MB,MDA and cTnI were decreased in group ER(P0.05).Conclusion Edaravone postconditioning combined with remote ischemic postconditioning can alleviate myocardial IR injury and offer better efficacy than either alone.
