Abstract

Objective To investigate the activation of β-tubulin after middle cerebral artery occlusion in the rats and treatment with Tongxinluo.Methods Ischemia was induced by temporary middle cerebral artery occlusion(MCAO).In ependyma and subependymal region,hippocampus dentate gyrus of rats treated with Tongxinluo,the numbers of β-tubulin were detected by immunohistochemistry at 3,7,14 and 21 days after MCAO.Results Compared with the sham operation group,β-tubulin was significantly increased in the MCAO on the 7,14 and 21 day(P0.05).Fluorescence intensity of BrdU+β-tubulin significantly increased in ependyma and subependymal region.After the treatment of Tongxinluo,the number of BrdU and fluorescence intensity of BrdU+β-tubulin were significantly increased as compared with the MCAO group(P0.05).Conclusions Focal cerebral ischemia in the rat results in a rapid response,a process often referred to as reactive β-tubulin,from resident astrocytes of the ependyma and subependymal region and hippocampus dentate gyrus to the side of ischemia;Tongxinluo enhances the ability of the differentiation and proliferation of neural stem cell in MCAO.
