Protective Effects of Salvianolic Acid B against Glucose Deprivation Induced Apoptosis of PC12 Cells

Abstract

Objective To study the protective effects of Salvianolic acid B(Sal B) against PC12 cells apoptosis induced by glucose deprivation.Methods The model of glucose deprivation was induced in PC12 cells.The cell survival rate was detected by methyl thiazolyl tetrazolium staining method.The apoptosis of propidium iodide stained PC12 cells was detected by flow cytometry.The expression of Caspase-3 in PC12 cells was detected by Western blotting method.Results Sal B could significantly inhibit glucose deprivation induced damage of PC12 cells in a dose-dependent manner within the concentration range from 0.1 to 100 μmol/ L.Flow cytometry showed that Sal B significantly decreased PC12 cells apoptosis induced by glucose deprivation.Western blotting showed that Sal B significantly decreased glucose deprivation-induced expression of Caspase-3.Conclusion Sal B has a protective effect against glucose deprivation-induced apoptosis of PC12 cells within the range of 0.1 to 100 μmol/ L,the protective mechanism of which may be related to Sal B inhibiting the expression of Caspase-3.
