Abstract
Objective To investigate the effect of different dose of heparin reduces acute lung injury in rats induced by Lipopolysaccharide (LPS).Methods LPS (5mg·kg-1) was infused by femoral vein to reproduce ALI model in rats.Fifty male SD rats were randomly divided into five groups with 10 animals in each group:in group C1-C3 LPS 5mg·kg-1 and heparin 250u·kg-1,500u·kg-1 and 1000u·kg-1 was infused by femoral vein simultaneously;group B received LPS 5 mg/kg;group C served as control one by giving the same volume of saline.At 4 h after LPS was given,and then the animals were killed.The pathological manifestation of lungs were observed,and the lung W/D ratio was calculated,and the concentration of malondialdehyde (MDA) in lung was detected,the concentration of TNF-a and protein in BALF were detected.Results Obvious ALI pathological manifestation in the lungs of B group rats were observed.Comparing with A group,the lung W/D ratio and concentration of TNF-a and protein in BALF significantly increased in B group (P0.01) as well as the concentration of MDA activity in lung (P0.01).Comparing with LPS group,the pathological manifestation in the lung and the lung W/D ratio and concentration of TNF-a and protein in BALF and the concentration MDA decreased in C1-C3 group (P0.05).However,bleeding complications increased with the dose of heparin increased.Conclution Early treatment of heparin can provide evident effects on ALI in rats,250u·kg-1 was the best treatment dose.
