Abstract
Objective:To investigate the effects of propofol and ischemic preconditioning on cell apoptosis induced by hepatic ischemia-reperfusion injury in rats. Methods: Seventy-two adult healthy SD rats were randomly divided into 4 groups(n=18).They were a sham-operated group(Sham group),an ischemia-reperfusion group(IR group),an ischemic preconditioning group(IP group)and a propofol group(P group).A 70% hepatic ischemia-reperfusion model was made and the left hepatic lobes were sampled after the experiment. The expression of Bcl-2 and Bax protein in liver tissue was assessed by immunohistochemical method and the apoptosis index(AI)of hepatic cells was measured by TUNEL method. Results: Compared with the Sham group, the expression of Bcl-2,Bax protein and the apoptosis index of hepatic cells in each group were significantly increased(P0.05).Compared with the IR group, the expression of Bcl-2 protein in the IP group and the P group was significantly increased while the expression of Bax protein and AI significantly decreased(P 0.05).Compared with the IP group, the expression of Bcl-2 protein in the P group was increased while the expression of Bax protein and AI decreased(P0.05).Conclusion: Propofol and ischemic preconditioning can relieve the liver cellular apoptosis by regulating the expression of Bcl-2 and Bax protein and may protect liver from ischemia-reperfusion injury in rats.
