Abstract
Objective To study the effect of Tiaogan Jiedu Formula(TJF) on learning and memory in rats with chronic epilepsy and expression of cAMP responsive element binding protein(CREB) in hippocampus.Methods Totally 60 chronic epilepsy model rats were randomly and equally divided into model group,TJF large-dose group,TJF small-dose group and Sodium Valproate group after intraperitoneal injection of Picrotoxin.The observation and treatment lasted for 60 days,and normal group of 15 rats were observed simultameously .The ability of learning and memory was tested by Morris water maze.The expression of CREB mRNA in hippocampus and level of phosphorylated CREB in hippocampus were detected by reverse transcription polymerase chain reaction and immunohistochemical method respectively.Results Memory ability,relative absorbance of CREB mRNA and positive expression of CREB immunocytes in TJF large-dose and small-dose groups increased significantly as compared with model group(P0.01 or P0.05).Conclusion TJF can improve learning and memory ability of rats with chronic epilepsy,which might be relevant to its regulating effect on the expression of CREB in hippocampus.
