Abstract
Objective To explore the fuction of transforming growth factor beta1(TGF-β1) in the progression of breast cancer and T lymphocyte transducted cellular immuno-reaction.Methods The effects of TGF-β1 on the preliferation of breastl cancer cell lines MCF-7,MDA-MB-157,SK-BR-3 and MDA-MB-453 were detected by MTT and the dosage-effect and time-effect relationship between cells and TGF-β1 were analyzed.Using enzyme-linked immumoadsorbent assay (ELISA),the production of IL-2 and IFN-γ in the presence or absence of TGF-β1 was measured.Results TGF-β1 could inhibit the growth of breast cancer cells with different effects at different concentrations only in the first three days.Under the regulation of TGF-β1,the concentration of IL-2 (296.5± 79.4) ng/L and IFN-γ (665.3± 107.0) ng/L in the supernatant of cultured PBMC was significantly lower than in the control group [(644.9± 105.5) ng/L and (922.3± 184.1) ng/L respectively, P 0.001].Conclusion TGF-β1 can inhibit the proliferation of breast cancer cells in a certain range of concentration and in a very short duration.It can inhibit the production of cytokines to depress the cellular immunization.
