Abstract
To establish quantitative analysis tetracycline class antibiotic residue of method to use solid-phase extraction-HPLC ultraviolet detection in beef.The residual tetracycline class antibiotic were determined in beef by using Waters 2695 high-performance liquid chromatography.The sample by 5% perchloric acid solution extraction,C18 solid phase extraction column purification,rotating enrichment,0.22μm microfiltration membrane filtration,reversed-phase chromatography separation,ultraviolet detection.chromatographic column for the Waters Nova-park C18 column(4 μm,3.9 mmi.d.×150 mm),with sodium dihydrogen phosphate solution-acetonitrile-methanol(volume ratio 70∶20∶10) as mobile phase,velocity for 0.8 mL/min,ultraviolet wavelength for 365 nm,each incoming sample 10 μL.The results showed that the oxytetracycline,tetracycline,aureomycin respectively detection limit of 0.0365 μg/mL,0.0430 μg/mL,0.0558 μg/mL, linear range for 0.1 μg/mL~1.0 μg/mL.Determination of 54 samples beef,only 1 samples beef aureomycin residue for 132 μg/kg,higher than China ministry of agriculture prescribed maximum permissible residues standard(100 μg/kg).In conclusion,the sale beef in lanzhou tetracycline antibiotic residues rate is not high(1/54).This article constructs solid-phase extraction-HPLC-ultraviolet assay determination beef tissues,such as oxytetracycline,tetracycline,aureomycin etc tetracycline class antibiotic residues are accurate and high accuracy.
