Abstract
[Objective] To investigate the roles of MDR, MRP in multidrug resistance of liver carcinoma cell lines. [Methods] To induce the multidrug resistance, human hepatic carcinoma cell lines were cultured in medium containing gradually increased concentratioins of ADM. MTT, FCM were applied to measure the resisance of cell line to chemical agents, the concentrations of chemical agents inside the cells; MDR, MRP expressions on the cell membrane, respectively. Verapamil was given to restore the sensitivity of cell lines to chemical agents. [Results] MDR, MRP markedly expressed on resistant cell lines HepG2/ADM, the intracellutar concentrations of chemical agents were significantly reduced; blocking the MDR, MRP functions with verapamil could restore the sensitivity of cell lines to chemical agents. [Conclusions] The high expressions of MDR, MRP were one of the important mechanisms of multidrug resistance of hepatic cell carcinoma.
