Experimental study on the effect of repairing and reconstruction by mesenchymal stem cells transplantation into the ischemic myocardium in rat

Abstract

Objective To investigate the proliferation and differentiation of mesenchymal stem cells(MSCs) when implanted into ischemic myocardium and the improvement of heart function. Methods SD rat MSCs were cultured in vitro and labeled with BrdU,then MSCs were transplanted into the ischemic myocardium whose left descending branch of coronary artey had been ligated(group Ⅰ),the rats receiving serum-free medium injection were used as controls(group Ⅱ). Four weeks after transplantation,MSCs's differentiation were observed. Echocardiography,SPECT,immunohistochemistry and angiogenesis were observed to detect the effects on heart function. Results Four weeks after transplantation of MSCs,the regeneration myocardial cells were observed in ischemic areas. Echocardiography indicated that the improvement of the LVEF value in group Ⅰ was significantly than that in grpup Ⅱ( P 0.05). SPECT demonstrated a significant revival of myocardial perfusion in the ischemic areas in group Ⅰ as compared with group Ⅱ( P 0.05). Conclusion MSCs can repair and reconstruct damaged myocardium and improve myocardial perfusion and heart function when transplanted into ischemic myocardium.
