Abstract
Bisphosphonates not only specifically inhibit osteoclasts,but also play a role in osteoblasts differentiation.Osteoblasts from neonatal rats were purified by enzyme digestion and cultured in vitro.Then they were divided into black contral group,high doses of lendronate,medium doses of lendronate and low doses of lendronate group.To observe the effect of lendronate on osteoblast differentiation by BMP-2/Smads/Runx2/Osterix signaling pathway,the results are as follows:after treatment 2 days and 4 days,the AKP level in medium dose of Alendronate group was significantly higher than that of the blank control group(p0.05).6 days after the intervention,but there was no statistical significance compared the treatment group with blank control group(p0.05).The results from ELASE showed that the BMP-2 level in the medium group obviously increased,(p0.05).The Collagen Type I level in treatment groups was significantly increased after 2 and 4 days(p 0.05).6 days after the intervention,although the Collagen Type I level osteoblasts in treatment groups decreased,it was still higher than that in black contral group(p0.05).The results from Realtime PCR showed that level of BMP-2,Smad1/5,Runx2 and Osterix mRNA in Alendronate groups was higher than that in the blank control group in the same period.So the conclusion is that Alendronate probably could stimulate osteoblast proliferation and differentiation through BMP-2/Smads/Runx2/Osterix signal conduction pathway.
