Abstract
Objective To study the molecular mechanism of Mycobacterium tuberculosis resistance to isoniazid (INH) and to establish a method of rapid detection of katG gene mutations in M.tuberculosis.Methods The katG genes of 105 M.tuberculosis clinical isolates were tested by PCR-RFLP and verified by DNA sequencing and bioinformatics analysis.Results Strain H37Rv was used as a control.In 105 M.tuberculosis clinical isolates,5 of 34 (14.7%) drug-susceptible isolates and 55 of 105 (52.4%) INH-resistant isolates had a katG gene mutation according to PCR-RFLP.Conclusion Drug resistance of M.tuberculosis to INH is related to katG gene mutation,PCR-RFLP might serve as a method of rapidly detecting the INH resistance of M.tuberculosis.
