Experimental study on the acute rejection of liver transplantation inhibited by pre-infusion of donor specific apoptosis cell

Abstract

Objective To investigate the effect and mechanism of acute rejection of liver transplantation inhibited by pre-infusion of donor specific apoptosis cell.Methods The spleen of SD rats were obtained under aseptic condition,then the apoptosis cells and necrotic cells were prepared by Photo luminescence method and water bath method,and confirmed by flow cytometry.Forty cases of SD rats(donor) and 40 cases of wistar rats(receptor) were devided into group A with 12 cases,group B with 14 cases and group C with 14 cases,then were injected with 1ml saline,1×107 necrotic cells and 1×107 apoptosis cells,7 days later the orthotopic liver transplantation were performed according to the Kamada method.The general conditions of the 3 groups during perioperative period were observed;Peripheral blood from all the groups were obtained on the fourth day after transplantation,and the ALT,AST,TBIL and serum IL-2,IL-10 were detected;Theree rats in each group were killed on the fourth day after transplantation,the liver tissue were obtained to study the pathological changes and decide the grade of acute rejection,the apoptotic index was detected through paraffin section;The splenic lymphocyte of SD rats and F344 rats were cultured with that of Wistar rats,the lymphocyte transformation rates were examined.Results The mean survival time of group C was significantly longer than that of group A and B,while the liver function,pathological changes and hepatocyte apoptotic index were better than those of group A and B,the lymphocyte transformation rates was significantly lower than that of group A and B.Conclusions The pre-infusion of donor specific apoptosis cell can effectively prevent acute rejection after liver transplantation by inducing the receptor specific immunological tolerance,also can improve the function of the transplanted liver and prolong survival time.
