Abstract
This experiment was designed to confirm that Hepatic Stimulator Substance (HSS).Insulin or/and Glucagon addition stimulated DNA synthesis(Hepatocyte Regeneration)in primary hepa-tocyte cell cultures prepared from adult rat liver and in human hepatoma cell line, Hep G2, in vitro. The results showed that HSS could strongly stimulate DNA synthesis in both of rat hepatocyte and hepatoma cells (Hep G2) (Compared with control, DNA synthesis increased 1. 14 and 1. 05 times, respectively. P0. 01). But no effect on fibroblast. However,Insulin or/and Glucagon at low concentration could not stimulate DNA synthesis in hepatocyte or Hep G2. High concentration of Insulin(10-5M) can moderately increase DNA synthesis (Compared with control 40% increase, P0. 05). Therefore,we concluded that HSS is a potential factor which can stimulate liver regeneration. Perhaps,it can be used in clinic. It may be better than the therapy of Insulin plus Glucagon which is widely used in clinic recently.
