Abstract

Objective To detect the expression of human β-defensin-2(hBD-2) mRNA induced by TNF-α,LPS,and IL-1β in the human airway primary epitheliums.Methods Human bronchial primary epitheliums were stimulated with TNF-α,LPS,and IL-1β and the expression of hBD-2 mRNA was detected by RT-PCR,Western Blot,and immunocytochemistry.Results hBD-2 mRNA could be detected in normal human primary epitheliums.The expression of hBD-2 mRNA was in a dose-dependent manner after stimula- tion with TNF-α,LPS,and IL-1β.hBD-2 could be detected at 3 h after stimulation with TNF-α(10 ng/ml), LPS(0.1 μg/ml),and IL-1 β(1 ng/ml).Conclusion LPS and proinflammarory factors can induce the expression of hBD-2 mRNA in a dose-dependent manner.The expression of hBD-2 may be an initial host de- fense against inflammatory stimulation in the airway.
