【Objectives】To study whether cyclooxygenase-2(COX-2) is overexpressed in CNE-1 Cell.To evaluate whether the selective inhibitor of COX-2(a selective inhibitor of COX-2) can inhibit the proliferation and induce the apoptosis of CNE-1 cell(a human nasopharyngeal carcinoma cell line).【Methods】 The protein expres-sion of COX-2 in CNE-2 cells was detected by immunohistochemical staining.The study was devived to four groups by the density of Celecoxib.The 0 μM,1.25 μM,2.50 μM,5.0 μM Celecoxib groups mean A,B,C,D group.MTT assay and colony forming assay were performed to determine the influence of Celecoxib on growth and proliferation of human nasopharyngeal cancer CNE-1 cells.Apoptosis of CNE-1 cells were determined by flow cytometry.【Result】The immunocytochemical staing showed that COX-2 was expressed in CNE-2 cell line cells.Celecoxib can inhibit the proliferation and colony forming and induce early and late stage apoptosis of CNE-1 cell line cells.There were differences in optical densities in MTT assay between group A and C and between group A and D,but no differences between group A and B.And there are differences between group B and C and between group C and D.These results indicate that Celecoxib has no effect on the proliferation of CNE-1 line cells at concentration 1.25 μM,but it has inhibitory effects at concentration of 2.50 μM and 5.00 μM,furthermore,inhibitory effects is more when the concentration of Celecoxib is larger.The numbers of colonies are lower and the early and late stage apoptosis cells are higher in Celecoxib alone group B,C and D,as compared with group A.Furthermore,there is difference in numbers of colonies and the early and late stage apoptosis cells between group B and D,but neither difference between group C and D nor between group B and D.【Conclusion】①CNE-1 cell line cells expressed COX-2.② Celecoxib can inhibit the proliferation and induce the apoptosis of CNE-1 cell line cells.③The anti-tumor effects of Celecoxib is more when the Celecoxib's concentration is larger at a range.
