Abstract

ObjectiveTo observe the effect of tetrandrine on the proliferation and expression of PCNA,CyclinD1 of hepatic stellate cell.MethodsHepatic stellate cells were cultured in RPM1640 medium.The effect of tetrandrine with various concentrations on the proliferation of hepatic stellate cell was observed with the MTT method.The effects of tetrandrine with various concentrations on the expression of PNCA,cyclinD1 of hepatic stellate cells were observed with immunocytochemistry.Results①From 0.5 mg/L to 1mg/L,tetrandrine inhibited the proliferation of hepatic stellate cell in a concentration dependence manner(P0.05).There was no significant difference between inhibition of 1mg/L tetrandrine and 2mg/L tetrandrine.② The number of the PCNA positive cells in 0.5mg/L,1mg/L tetrandrine groups were less than that of normal control group(P0.01),and the number of the PCNA positive cells had significant difference between 0.5mg/L and 1mg/L tetrandrine groups(P0.01).③The number of the CyclinD1 positive cells in 0.5mg/L,1mg/L tetrandrine groups were less than that of normal control group(P0.01),and the number of the CyclinD1 positive cells had significant difference between 0.5mg/L and1mg/L tetrandrine groups(P0.01).ConclusionTetrandrine can decrease the expression of PCNA and CyclinD1 of hepatic stellate cells,and make cell cycle arrest in G0/G1 stage,which may be the mechanism that tetrandrine inhibits the proliferation of hepatic stellate cell.
